چکیده مقاله:

Background: Allergy is a clinical disorder affecting the human population with wide geographical distribution. Platanus orientalis (P. orientalis) trees are planted in many countries and their pollen causes allergic reactions. Cyclophilin has recently been identified as one of the most important allergens of P. orientalis pollen. We aimed to clone and purify this allergen in Escherichia coli for further studies and therapeutic and diagnostic purposes for allergy to P. orientalis. Methods: RNA was extracted from P. orientalis. A full-length fragment encoding cyclophilin was prepared by polymerase chain reaction amplification of the first-strand cDNA synthesized from P. orientalis RNA. The cDNA was inserted into the pET32b (+) vector, and the construct transformed into E. coli Top10 and BL21 cells. The expressed protein was purified by the CuSO4 method. Results: The cDNA for the cyclophilin of P. orientalis pollen was cloned, and a specific reactivity of recombinant cyclophin was confirmed by immunoblotting using sera from patients allergic to P. orientalis pollen. Conclusion: The recombinant cyclophilin has a potential for immunologic assays for evaluation of allergy to P. orientalis pollen.

Cloning and Expression of Mannheimia haemolytica PlpE Gene in Escherichia coli and its Immunogenicity Assessment

Mannheimia haemolytica is responsible for considerable economic losses to cattle, sheep, and goat industries in many parts of the world. This bacterium isone of the causative agents of shipping fever in cattle. Current vaccines against M. haemolytica are moderately efficacious since they do not provide complete protection against the disease. Production of a...

Cloning and expression of Eimeria necatrix microneme5 gene in Escherichia coli

Background: Coccidiosis caused by Eimeria necatrix has the most economic impact onpoultry production. Micronemal proteins in Eimeria necatrix are thoughtto be critical ligands determining host cell specificity at the time ofinvasion.               OBJECTIVES: Isolation and purification of Eimeria necatrix oocysts from  Khuzestan province of Iran was performed. AcDNA encoding microneme 5 (EnMIC5...

cloning and expression of functional reteplase in escherichia coli top10

background: production of tissue plasminogen activator protein (t-pa) in prokaryotes systems has many problems such as the lack of active protein production, multiple purification steps, and renaturation process which has been shown to be costly and time-consuming. methods: in this study, reteplase which is the nonglycosylated active domain of t-pa was used to transform top10 escherichia coli (...

Cloning and Expression of Influenza H1N1 NS1 Protein in Escherichia Coli BL21

Background:Influenza virus is globally pathogenic and it is usually associated with zoonotic respiratory diseases. This virus has caused a number of pandemics with a high mortality rate. The non-structural (NS1) protein of influenza A viruses is a non-essential virulence factor that has multiple accessory functions during viral infection. This protein is highly conservative. It has been shown t...
